Principle of the Assay: The Human SAA1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human SAA1 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Human SAA1 coated on a 96-well plate. Standards and samples are pipetted into the wells and SAA1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human SAA1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of SAA1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: Serum amyloid A protein (SAA), also known as SAA1, is a protein that in humans is encoded by the SAA1 gene. This gene encodes a member of the serum amyloid A family of apolipoproteins. It is mapped to 11p15.1. SAA is a major acute phase protein that is highly expressed in response to inflammation and tissue injury. This protein also plays an important role in HDL metabolism and cholesterol homeostasis. High levels of this protein are associated with chronic inflammatory diseases including atherosclerosis, rheumatoid arthritis, Alzheimer's disease and Crohn's disease. This protein may also be a potential biomarker for certain tumors
